โมเลกุลส่งสัญญาณที่สอง

มีระบบโมเลกุลส่งสัญญาณที่สองหลายอย่าง (เช่น cAMP, phosphoinositol, และ arachidonic acid) แต่ทั้งหมดก็มีกลไกทั่วไปที่คล้ายกันมาก แม้สารที่เกี่ยวข้องและผลที่ได้จะต่างกัน

โดยมาก จะมีลิแกนด์หนึ่งที่จับกับโมเลกุลโปรตีนตัวรับ (receptor protein) ซึ่งแผ่ข้ามเยื่อหุ้มเซลล์ นอกเซลล์ การจับกันระหว่างลิแกนด์กับโปรตีนตัวรับทำให้ตัวรับแปรสภาพแบบ conformational change ซึ่งมีผลต่อการทำงานของตัวรับและมีผลให้สร้างโมเลกุลส่งสัญญาณที่สอง

ในกรณีของ G protein-coupled receptors การแปรสภาพจะเปิดจุดยึด (binding site) ของโปรตีนตัวรับกับจีโปรตีน (G-protein ตั้งชื่อตามโมเลกุลที่เข้ายึดกับโปรตีนคือ guanosine diphosphate ตัวย่อ GDP และ guanosine triphosphate ตัวย่อ GTP) ซึ่งยึดอยู่ที่เยื่อหุ้มเซลล์ด้านในโดยมีหน่วยย่อย ๆ 3 หน่วยคือ แอลฟา บีตา และแกมมา จีโปรตีนเรียกอีกอย่างได้ว่า ตัวแปรสัญญาณ (transducer)
