เส้นทางสายไหม (อังกฤษ: Silk Road หรือ Silk Route) เป็นชุดเส้นทางการส่งการค้าและวัฒนธรรมซึ่งเป็นศูนย์กลางของอันตรกิริยาทางวัฒนธรรมผ่านภูมิภาคของทวีปเอเชียที่เชื่อมตะวันตกและตะวันออกโดยการโยงพ่อค้าวาณิช ผู้แสวงบุญ นักบวช ทหาร ชนเร่ร่อนและผู้อาศัยในเมืองจากจีนและอินเดียไปยังทะเลเมดิเตอร์เรเนียนระหว่างเวลาหลายสมัย

เส้นทางสายไหมมีความยาว 6,437 กิโลเมตร (4,000 ไมล์) ได้ชื่อมาจากการค้าผ้าไหมจีนที่มีกำไรมากตลอดเส้นทาง เริ่มตั้งแต่ราชวงศ์ฮั่น (206 ปีก่อน ค.ศ. – ค.ศ. 220) ราชวงศ์ฮั่นขยายเส้นทางการค้าส่วนเอเชียกลางราว 114 ปีก่อน ค.ศ. ส่วนใหญ่ผ่านคณะทูตและการสำรวจของผู้แทนทางการทูตจักรวรรดิจีน จางเชียน (Zhang Qian) ชาวจีนสนใจมากกับความปลอดภัยของผลิตถัณฑ์การค้าของพวกตนและขยายกำแพงเมืองจีนเพื่อประกันการคุ้มครองเส้นทางการค้านี้

การค้าบนเส้นทางสายไหมเป็นปัจจัยสำคัญต่อการพัฒนาของอารยธรรมจีน อนุทวีปอินเดีย เปอร์เซีย ทวีปยุโรปและคาบสมุทรอาหรับ โดยเปิดอันตรกิริยาทางการเมืองและเศรษฐกิจทางไกลระหว่างอารยธรรม แม้ผ้าไหมเป็นสินค้าหลักจากจีน แต่ก็มีการค้าสินค้าอื่นจำนวนมาก ศาสนา ปรัชญาหลายความเชื่อและเทคโนโลยีต่าง ๆ จนถึงโรคก็ไปมาตามเส้นทางสายไหมเช่นกัน นอกเหนือจากการค้าทางเศรษฐกิจแล้ว เส้นทางสายไหมยังใช้เป็นการค้าทางวัฒนธรรมในบรรดาอารยธรรมตามเครือข่ายเส้นทางด้วย

ผู้ค้าหลักระหว่างยุคโบราณ คือ ชาวจีน เปอร์เซีย กรีก ซีเรีย โรมัน อาร์มีเนีย อินเดียและแบกเตรีย (Bactrian) และตั้งแต่คริสต์ศตวรรษที่ 5 ถึง 8 เป็นชาวซอกเดีย (Sogdian) ระหว่างการเจริญของศาสนาอิสลาม พ่อค้าอาหรับกลายมาโดดเด่น

ในการประชุมคณะกรรมการมรดกโลกสมัยสามัญครั้งที่ 38 พ.ศ. 2557 ณ กรุงโดฮา ประเทศกาตาร์ มีมติให้ขึ้นทะเบียนเส้นทางสายไหมในจีน คาซัคสถาน และคีร์กีซสถาน เป็นมรดกโลกภายใต้ชื่อ เส้นทางสายไหม : โครงข่ายเส้นทางฉนวนฉางอาน-เทียนชาน โดยให้เหตุผลว่า เส้นทางนี้เป็นที่ยอมรับกันว่ามรดกทางอารยธรรของมนุษยชาติ ในฐานะเป็นเส้นทางโบราณในการติดต่อค้าขายและสื่อสารระหว่างตะวันออกกับตะวันตก
