ปลาฉลามมาโก (อังกฤษ : Mako shark ) ปลากระดูกอ่อนจำพวกปลาฉลาม ในสกุล Isurus ปลาฉลามขาว ซึ่งเป็นปลากินเนื้อและสัตว์กินเนื้อที่ใหญ่ที่สุดในโลกที่ยังคงสืบทอดเผ่าพันธุ์มาจนถึงปัจจุบัน
ปลาฉลามมาโก จัดเป็นปลาฉลามที่มีความปราดเปรียวว่องไว เป็นปลาฉลามที่ว่ายน้ำได้เร็วที่สุดในโลก โดยทำความเร็วได้ถึง 50-70 กิโลเมตรต่อชั่วโมง ทั้งนี้เป็นผลมาจากการขยับหางที่เว้าเป็นรูปพระจันทร์เสี้ยวขนาดใหญ่ ซึ่งสามารถเพิ่มความเร็วได้ในช่วงระยะเวลาเสี้ยววินาที เพื่อใช้ในการล่าเหยื่อประเภทต่าง ๆ เช่น โลมา, แมวน้ำ หรือเต่าทะล จากการจู่โจมจากด้านล่าง อีกทั้งยังสามารถกระโดดได้พ้นน้ำได้สูงถึง 9 เมตรอีกด้วย เป็นปลาที่มีดวงตากลมโตสีดำสนิท ภายในปากมีฟันแหลมคมจำนวนมากจนล้นออกนอกปาก จัดเป็นปลาฉลามอีกจำพวกหนึ่งที่ทำอันตรายมนุษย์ได้
เป็นปลาที่ถือกำเนิดมาจากตั้งแต่ยุคครีเตเชียส จนถึงควอเทอนารี (ประมาณ: 99.7 ถึง 0.781 ล้านปีก่อน)
จำแนกออกได้เป็น 2 ชนิด ที่ยังคงสืบทอดเผ่าพันธุ์มาจนถึงปัจจุบัน มีความยาวประมาณ 2.5 ถึง 4.5 เมตร (8.2 ถึง 14.8 ฟุต) และน้ำหนักมากที่สุดถึง 800 กิโลกรัม (1,800 ปอนด์)
โดยคำว่า "มาโก" มาจากภาษามาวรี หมายถึง ปลาฉลามหรือฟันปลาฉลาม
